I have made a very large Web Help project and the client has asked to change the color of the lines and text in the screen shot annotations. Do I have to do this image by image, or is there a batch way to do this?

Welcome to the forums.
If you are trying to change content within your help topic you will need to find how the styla has been applied.
Is the style inline? If so you will have to do a search (and replace) across the project to find each instance of the style. If you have everything in a stylesheet, it will be a lot easier. Change the relevent properties in your stylesheet and away you go.
If it's the image itself, its a different story.
